The Essex opener lined up alongside Joe Denly in the absence of Andrew Strauss at the top of the order, with England put into bat by Proteas skipper Graeme Smith.
With regular captain Strauss unavailable due to his decision to no longer play international 20-over cricket, Paul Collingwood led the tourists, who also included fit-again James Anderson in the team.
But there was no place for Graham Onions despite his recovery from lower back trouble.
Ryan McLaren made his debut for the hosts, while Loots Bosman came in to open for the first time in more than a year.
